MGI Upgrades DNBSEQ-G400 Gene Sequencer to be 10 Hours Faster

Recently, MGI has fully upgraded its medium-to-high throughput gene sequencer DNBSEQ-G400 to provide more stable performance and better user experience. As a versatile desktop sequencer, DNBSEQ-G400 can provide users with comprehensive, flexible and efficient sequencing options, and it is the preferred model for medium-to-high throughput sequencing laboratories.



Since the release, DNBSEQ-G400 has been widely used in scientific research, disease control and prevention, environment, and agriculture. More than 560 papers have been published in high-quality science journals. MGI is committed to developing life science tool and technology for precision medicine, precision agriculture, precision healthcare and other relevant industries. MGI has recently upgraded the control software, algorithm software and supporting scripts of DNBSEQ-G400, bringing customers faster, simpler, and more flexible sequencing experience!


Speed up by 20% with Faster, Larger Flow Cell

With the PE150 sequencing mode, DNBSEQ-G400 can generate >1 Tb base data by running 2 FCL Cells. DNBSEQ-G400 has now been upgraded with a new version of sequencing reagents to speed up the process! While guaranteeing both quality and quantity, the run time of FCL Flow Cell at PE100 is shortened from 48h to 38h, and FCL Flow Cell at PE150 from the original 66h to 56h, about 20% faster respectively.

Through algorithm optimization, DNBSEQ-G400 is friendlier to base imbalance libraries, and even a single barcode can achieve good splitting. As shown in Figure 2, the NA12878 sample is used for PCR-Free WGS library building, and the sequencing process is conducted on the upgraded DNBSEQ-G400. The data shows that the SplitRate of both single-barcode samples and two-barcode samples reach more than 98%, satisfying flexible sequencing and splitting needs of small sample size.

Methylation library is a typical imbalance library. In order to achieve more accurate base signal identification during sequencing, it is necessary to add a certain proportion of balance library for base balance. After DNBSEQ-G400 is upgraded, the split rate of WGBS sequencing can reach 96% when it is performed without adding a balance library, which is nearly 60% higher than that of before upgrading (60.39%), as shown in Figure 3. From the analysis of the sequencing performance of adding 25% of balance library and without adding balance library, it can be seen that adding balance library or not has no significant effect on the amount of data generated by DNBSEQ-G400 sequencing (Figure 4).

More Flexible with Dual-Mode Sequencing, Ready to Run at Any Time


After the upgrade, it can realize dual-mode sequencing, which means the AB side can be sequenced independently with arbitrary interval time. A second slide can be added in the middle of the sequencing for dual-slide operation mode. The two slides run independently, and the sequencing data can be output independently to further shorten the turn-around time, so that there is no need to wait for sequencing. Samples can be sequenced at any time. One instrument can work as “two” to greatly improve the use efficiency of the sequencer.
