MGISP-100: Compatible with Multiple Sequencing Libraries, Enabling Automated Library Conversion

MGISP-100 Automated Sample Preparation System enables high-throughput sequencing library preparation by integrating all library preparation functions in one. It now supports automated operation of more than 40 MGI and third-party library preparation kits. With MGISP-100 Automated Sample Preparation System, MGIEasy Universal Library Conversion Kit (App-A) is designed for conversion from the linear dsDNA library prepared by the third-party library kit to the single stranded circular (ssCir) DNA library compatible with MGI's sequencing platform using only two steps: library adapter conversion and sequencing primer replacement. Together with the automated Make DNB process, DNA nanoballs (DNBs) can be directly sequenced on the DNBSEQ™ platform.




The unique Rolling Circle Amplification (RCA) technology of the DNBSEQ™ platform effectively avoids exponential amplification of errors compared with exponential polymerase chain reaction (PCR) amplification, while featuring low index hopping and low amplification bias. The preparation of the unique circular library with the DNBSEQ™ platform requires multiple steps including enzyme cleavage, end repair, adapter ligation, product purification, PCR amplification, and circular DNA digestion. The DNBSEQ™ library, which was prepared on MGI's MGISP automated platform, can significantly shorten experimental time and improve stability of library preparation, compared with manual operation.


The automation scripts of MGI App-A+Make DNB running on MGISP-100 has been tested strictly and repeatedly, which maximizes the stability and repeatability of automated library preparation, and, at the same time, realizes the compatibility of non-DNBSEQ™ libraries with the DNBSEQ™ sequencing platform.


Automated library conversion on MGISP-100 mainly includes four automation steps, namely PCR library preparation, automated pooling, circular DNA digestion, and Make DNB. It can support up to 16 samples at a time, and the highly automated process only takes 4.5-5 hours in total. During this period, the operator only needs to do some simple quality control and monitoring work, which takes about 0.5 hours. If the whole process is manually operated, it could take more than 2 hours for the operator to process a single sample or multiple samples.


The results show that MGISP-100 Automated Sample Preparation System is on par with or even better than manual operation in terms of AC-PCR products and the total amount of libraries prepared. MGISP-100 and APP series can quickly and effectively enable the automatic, non-destructive conversion of a batch of linear libraries into libraries compatible with DNBSEQ™ where high-throughput sequencing is available, thus providing linear library users with more sequencing options, which greatly improves the adaptability and compatibility of the sequencing platform and validates the versatility and stability of MGISP series library preparation systems.
